Both diseases sharing common features, we developed a mechano-chemical model of the parenchyma to investigate tissue destruction, initially based on LAM and subsequently extended to COPD. In both diseases, tissue destruction is driven by proteases release due to complex interactions between cells and proteins during injury and abnormal repair processes. This impacts collagen and elastin density in lung tissues, potentially resulting in alveolar wall thinning. To investigate these mechanisms, we model tissue integrity using a system of coupled ordinary differential equations. Simultaneously, we assess the mechanical strains experienced by the springs in the network. As tissue integrity decreases, any increase in strains exceeding a physiological threshold eventually leads to springs breaking (i.e., tissue damage). Our model allows us to study the mechanisms underlying tissue destruction in LAM and COPD and to investigate time-dependent network-level measures from which potential biomarkers could be derived. Coupled with clinical images and spatial omics data, it could help clinicians predict the evolution of both diseases and help develop optimal treatments.
